摘 要: 目的 通过对煤工尘肺(CWP)患者血清中8项生物标记物水平测定及因子分析,为探讨CWP发生机制提供新的依据。方法 采用酶联免疫吸附法检测116例CWP患者血清中IL-18、IL-18R、IL-8、s CD40、s CD40L、MCP-1、MMP-9及TIMP-9等8项指标水平,采用SPSS17.0软件对其进行因子分析。结果 CWP组与对照组比较,IL-18、IL-18R、IL-8、s CD40L和TIMP-9水平升高,差异均具有统计学意义(P<0.01),而s CD40、MCP-1和MMP-9水平则无明显改变(P>0.05)。8项原始指标信息量提取率最高者为93.3%,最低者为64.1%;经因子分析提取到4个公因子,分别为炎症因子(F1)、共刺激分子(F2)、基质金属蛋白酶及其组织抑制因子(F3)、细胞因子受体(F4),其方差贡献率分别为33.06%、21.60%、14.16%、13.43%,累计方差贡献率为82.25%。结论 CWP患者血清8项生物标记物相关因素依次为炎症因子、共刺激分子、基质金属蛋白酶与其组织抑制因子和细胞因子受体等4种,因子分析法可能对今后探讨CWP发病机制、治疗等起指导作用。 |

Factor analysis on eight serum biomarkers in patients with coal worker's pneumoconiosis |

Abstract: Objective The aim of this study is to analyze eight serum biomarkers by factor analysis technique to offer some new theoretic basis for exploring the pathogenesis of coal worker's pneumoconiosis (CWP). Methods The ELISA method was used to measure the levels of IL-18,IL-18R,IL-8,sCD40,sCD40L,MCP-1,MMP-9 and TIMP-9 in serum of 116 CWP patients,and SPSS17.0 software was used for factor analysis. Results The serum levels of IL-18,IL-18R,IL-8,sCD40L and TIMP-9 were significantly higher in CWP compared with control group (all P<0.01),while the serum levels of sCD40,MCP-1 and MMP-9 showed no obvious change (all P>0.05). The maximum extraction rate was 93.3%,the minimum was only 64.1% in these eight initial components; four comm factors extracted were as follows: inflammatory factors(F1),costimulatory molecules (F2),matrix metalloproteinase and its tissue inhibitors (F3) and cytokine receptors (F4),the variance contribution rates were 33.06%,21.60%,14.16% and 13.43% respectively, cumulative variance contribution rate was 82.25%.Conclusion The study showed that there were four pathogenic routes involving serum biomarkers in CWP: inflammatory factors,costimulatory molecules,matrix metalloproteinase and its tissue inhibitors,cytokine receptors,suggested that factor analysis should be helpful in exploration on the pathogenesis and treatment of CWP. |
