摘 要: 目的 研究双氢青蒿素对染矽尘大鼠肺纤维化的干预作用,比较双氢青蒿素和汉防己甲素对染矽尘大鼠肺纤维化的干预效果。方法 将80只SD大鼠随机分成对照组、模型组、双氢青蒿素组、汉防己甲素组,每组20只。对照组大鼠气管内注入生理盐水1 ml,其余3组大鼠气管内注入浓度为50 mg/ml 的二氧化硅混悬液1 ml染尘,染尘后第2天开始灌胃给药。双氢青蒿素组给予双氢青蒿素75mg/kg,汉防己甲素组给予汉防己甲素22 mg/kg,模型组和对照组给予生理盐水1 ml/100 g,每周给药6 d,连续给药3个月,并于染尘后的第7、14、28、90天分批处死大鼠,取大鼠肺组织,检测肺组织羟脯氨酸含量,观察肺组织病理学改变,酶联免疫法(Elisa)检测大鼠血清中转化生长因子-b1(TGF-b1)和Smad2/3含量。结果 双氢青蒿素、汉防己甲素和模型组羟脯氨酸含量均高于对照组(P<0.05),双氢青蒿素、汉防己甲素组羟脯氨酸含量低于模型组(P<0.05);双氢青蒿素和汉防己甲素组之间羟脯氨酸含量差异无统计学意义(P>0.05)。双氢青蒿素和汉防己甲素组大鼠血清中TGF-b1、Smad2/3表达水平均低于模型组(P<0.05),双氢青蒿素和汉防己甲素组之间大鼠血清TGF-b1与Smad2/3表达水平比较差异无统计学意义(P>0.05)。结论 双氢青蒿素对染矽尘大鼠肺纤维化有抑制作用,双氢青蒿素和汉防己甲素对染矽尘大鼠肺纤维化的干预效果无明显差异。 |

Intervention effect of dihydroartemisinin on pulmonary fibrosis by silica exposure in rats |

Abstract: Objective To study the intervention effect of dihydroartemisinin, and compare the effect with tetrandrine on pulmonary fibrosis in rats exposed to silica. Methods A total of 80 SD rats were randomly divided into 4 groups:control group, model group, dihydroartemisinin group and tetrandrine group, 20 rats in each group. Rats of control group were intratracheally injected with 1 ml of normal saline, the other three groups were injected with 1ml of silica suspension at a concentration of 50 mg/ml; drugs were administered intragastrically at the next day after silica exposur:75mg/kg of dihydroartemisinin, and 22 mg/kg of tetrandrine, respectively, 6 times/week for 3months (the model group and the control group were given 1ml/100g of normal saline). Then the rats were killed in batches on the 7th day, 14th day, 28th day, and 90th day after silica exposure respectively, and the lung tissues of the rats were taken at the same time, the hydroxyproline content, and the pathological changes of lung tissue were detected, the levels of transforming growth factor-1 (TGF-1) and Smad2/3 in serum were measured by enzyme-linked immunosorbent assay (Elisa). Results The results showed that the contents of hydroxyproline in dihydroartemisinin, tetrandrine and model groups were all higher than those of control group (P0.05), while the levels of hydroxyproline in dihydroartemisinin and tetrandrine group were all lower than that of model group (P0.05), there was no significant difference in hydroxyproline level between dihydroartemisinin and tetrandrine groups (P0.05). The serum levels of TGF-1 and Smad2/3 of dihydroartemisinin and tetrandrine group were lower than those in model group, (P0.05), but there was no significant difference in the expression of serum TGF-1 and Smad2/3 between the dihydroartemisinin and tetrandrine groups (P0.05). Conclusion The results suggested that dihydroartemisinin had an inhibitory effect on pulmonary fibrosis in rats exposed to silica, there was no significant difference in the intervention effect between dihydroartemisinin and tetrandrine on pulmonary fibrosis in rats exposed to silica. |
