摘 要: 目的 研究中药颗粒对染矽尘、 Col-Ⅰ的影响, 并从 TGF-β1 / Smads 通路探讨其可能的作用机制。 方法将 SD 雄性大鼠随机分为对照组、 模型组和中药高、 低剂量组, 每组 16 只。 对照组大鼠气管灌注 1. 0 ml 生理盐水,其余组大鼠均给予 1. 0 ml 5%无菌 SiO2悬浊液; 造模后第 2 天开始给大鼠灌胃给药, 1 次/ d, 高剂量组、 低剂量组大鼠分别灌胃 1 000 mg / kg、 500 mg / kg 中药, 对照组、 模型组给予 0. 9%生理盐水。 于灌胃第 14、 28 天处死大鼠, 并检测大鼠肺组织Col-Ⅰ、 TGF-β1 的含量及 TGF-β1、 Smad2 / 3 蛋白的表达水平。 结果 造模后第 14 天和第 28 天, 模型组大鼠肺组织Col-Ⅰ、 TGF-β1 含量高于对照组, 中药干预后Col-Ⅰ、 TGF-β1 含量有所降低, 不同剂量组间差异均具有统计学意义 (P<0. 05); 模型组大鼠肺组织 TGF-β1、 Smad2 / 3 及 p-Smad2 / 3 蛋白的表达水平高于对照组, 中药干预后TGF-β1、 Smad2 / 3 及 p-Smad2 / 3 蛋白的表达水平有所降低, 不同剂量组间差异也具有统计学意义 (P<0. 05)。 结论中药颗粒可以抑制 SiO2诱导的大鼠肺组织Col-Ⅰ含量的升高, 且该抑制作用可能与中药颗粒对大鼠肺组织 TGF-β1 /Smads 通路的调节有关。 |

Regulatory effect of Chinese herbal compound on TGF-β1 / Smads in lung tissues of rats exposed to silica dusts |

Abstract: Objective To study the effect of traditional Chinese medicine (TCM) particles on Col-Ⅰ in lung tissues of rats exposed to silica dusts and explore its possible mechanism by TGF-β1 / Smads pathway. Methods SD male rats were randomly divided into control group, model group, high-dose group and low-dose group, each group had 16 rats; the control rats were intratracheally perfused with 1. 0 ml of normal saline, the rats in other groups were perfused with 1. 0 ml 5% SiO2 aseptic suspension. From the next day after silica exposure, the TCM was given by irrigation once a day, rats in high-dose group and low-dose group were given 1 000 mg / kg and 500 mg / kg TCM particles, respectively, the control rats and model rats were administered with 0. 9% saline. Then, the rats were killed at 14th and 28th day after intragastric administration, respectively,and to detect the contents of Col-Ⅰ and TGF-β1, and the proteins expression levels of TGF-β1 and Smad2 / 3 in lung tissues.Results Both the 14th day and the 28th day after silica exposure, the contents of Col-Ⅰ and TGF-β1 in rats lung tissues of model group were all higher than those of control group, all of them were dose-dependently decreased after intervention of traditional Chinese medicine, the difference was statistically significant (P<0. 05); meanwhile, on the 14th day and the 28th day after silica exposure, the expression levels of TGF-β1 and Smad2 / 3 protein in lung tissues of model group were higher than those of control group, the expression level of TGF-β1 and Smad2 / 3 protein was also dose-dependently decreased after intervention of traditional Chinese medicine, the difference was statistically significant ( P< 0. 05). Conclusion TCM particles for fibrosis could inhibit the increase of Col-Ⅰ in SiO2-induced lung tissues of rats, the mechanism may be related to the effect on TGF-β1 /Smads pathway in lung tissues of rats. |
