摘 要: 目的 验证中药黄芩苷的抗氧化以及对油酸所致急性呼吸窘迫综合征(ARDS)氧合功能和组织损伤的改善作用。方法 将雄性SD大鼠随机分为对照组、油酸组和黄芩苷低、中、高剂量(150、300、450 mg/kg)组,在10 min、1 h、2 h、6 h不同时间点采集动脉血测定氧分压(PaO2);根据实验结果选取某一剂量黄芩苷组作为治疗组,与对照组、油酸组进一步实验,在10 min、30 min、1 h、2 h、6 h、12 h和24 h时测定PaO2,同时检测血常规、动脉血气分析、肺组织内髓过氧化物酶(MPO)和活性氧自由基(ROS)等指标,并进行组织病理学检查。结果 2 h时油酸组的氧合指数达到最低值(同对照组相比,P<0.01),为189.45±43.03mmHg,符合ARDS的诊断(<200 mmHg);黄芩苷中剂量组氧合指数改善最为明显,故选取该剂量组作为治疗组。进一步的实验结果显示,油酸组WBC、中性粒细胞计数(N)和百分比计数与对照组相比有统计学意义,治疗组这些指标出现峰值的时间晚于油酸组;油酸组和治疗组肺组织ROS含量均高于对照组,但治疗组峰值低于油酸组,且6 h时已接近对照组;MPO浓度具相同的变化规律。HE染色和病理评分显示,治疗组肺损伤程度较油酸组轻。结论 适宜剂量的黄芩苷通过减少中性粒细胞在肺内的集聚和ROS的产生发挥抗氧化作用,进而改善ARDS的缺氧状态。 |

Antioxidative Effects of Baicalin on oleic acid induced-acute respiratory distress syndrome in rats |

Abstract: Objective To investigate the antioxidative function of baicalin,which can improve the anoxia status and tissue damage of on oleic acid induced-acute respiratory distress syndrome (ARDS). Methods Healthy adult male SD rats were divided into five groups: control group, oleic acid group and baicalin low, medium and high dose (150, 300, 450 mg/kg) groups. Arterial blood was collected at different time points of 10 min, 1 h, 2 h and 6 h to determine the partial pressure of oxygen (PaO2). According to the experimental results, baicalin group with a certain dose was selected as the treatment group。The next experiment was carried out with treatment group, control group and oleic acid group. Observe the changes in PaO2 at 10 min, 30 min, 1 h, 2 h, 6 h, 12 h and 24 h. In the meantime, changes of White blood cell count, neutrophil count, pulmonary myeloperoxidase (MPO) and reactive oxygen species (ROS), histopathological examination were performed too. Results The level of PaO2 and Oxygenation index showed decrease (P<0.01 vs. control) and come up to the diagnostic standard of ARDS at 2 h [Oxygenation index was (189.45±43.03) mmHg] in oleic acid group. The improvement of oxygenation index was most obvious in baicalin medium dose group, so this dose group was selected as the treatment group. In the subsequent experiments, Compared with the control group, WBC, neutrophil count (N) and percentage count in the oleic acid group were statistically significant compared with those in the control group, and the peak time of these indexes in the treatment group was later than that in the oleic acid group。The ROS content in lung tissue of oleic acid group and treatment group was higher than that of control group, but the peak value of ROS in treatment group was lower than that in oleic acid group, and was close to control group at 6 h. The change pattern of MPO concentration has the same. HE staining and pathological score showed that the degree of lung injury in the treatment group was less than that in the oleic acid group. Conclusion Appropriate dose of baicalin has good antioxidative effect, can improve hypoxemia of oleic acid induced-ARDS in rats, which can be related to inhibition of neutrophil aggregation and ROS production. |
