摘 要: 研究邻苯二甲酸二乙基己酯[BF](DEHP)及镉(Cd)对大鼠睾丸巨噬细胞(TM)[BFQB]分泌细胞因子IL-1β的影响。 方法 利用贴壁法提取大鼠睾丸巨噬细胞。以0.1%二甲基亚砜(DMSO)为对照组,用50 μg/ml脂多糖(LPS)作为激活剂,以1 μmol/L、10 μmol/L、100 μmol/L的DEHP或CdCl2单独及各自半量联合处理,作用24 h后用ELISA法检测上清液中细胞因子IL-1β的含量;用CCK8法测定各浓度DEHP或CdCl2单独处理对细胞活性的影响。 结果 LPS可明显促进巨噬细胞分泌细胞因子IL -1β;0.5 μmol/L DEHP+0.5 mol/L Cd和5 μmol/L DEHP+5 μmol/L Cd联合染毒可显著抑制细胞分泌功能,但IL-1β分泌量仍明显高于对照组,并介于两单独作用组之间;50 μmol/L DEHP+50 μmol/L Cd的联合处理可非常显著抑制细胞分泌功能,与对照组相比差异无统计学意义。随着剂量增加,DEHP或Cd单独作用都能抑制细胞分泌功能和细胞活性,但对两者抑制程度存在较大差异。 结论 DEHP和Cd联合处理抑制大鼠TM分泌IL-1β表现出一定交互作用,对细胞活性的抑制可部分解释DEHP和Cd对细胞分泌功能的抑制效应。 |

Abstract: To study the effect of di-2-ethylhexylphthate(DEHP)and cadmium(Cd)on secretion of a cytokine IL-1β by testicular macrophages(TM)in rats. Methods Attachment culture was used to extract TM of rats, 0.1% dimethyl sulfoxide(DMSO)as control and 50 μg/ml lipopoly saccharide (LPS )as activator,cells were exposed to different concentrations of DEHP,CdCl2 respectively or their mixtures(half dosage)for 24 hours.IL-1β in cell supernatants were determined by ELISA,and cell viability was detected by CCK8 method as well. Results The results showed that LPS obviously promoted the secretion of IL-1β by TM;both DEHP and Cd decreased the level of IL-1β by TM and the cell activity significantly,especially the former;cell secretion function could be also obviously inhibited by the mixtures;and effect of DEHP,Cd or the mixture on IL-1β secretion was dose dependent.[WTHZ]Conclusion The inhibition of DEHP and Cd on IL-1β secretion of TM had some interaction effect ,the suppression of cell viability can partialy interpret the decrease of IL-1β secretion by DEHP and Cd in TM |
