摘 要: 目的 探索60Co γ射线全身照射后大鼠尿液中硫氧还蛋白(Trx)、再生胰岛来源3 γ蛋白(REG3G)和锌-α2糖蛋白(AZGP1)表达水平变化。方法 30只Sprague Dawley(SD)大鼠随机分为0、0.5、1、3、5、8 Gy共6组,每组5只,给予60Co γ射线照射(剂量率1.0 Gy/min),收集照射后1 d、3 d、5 d大鼠尿液,用ELISA方法定量检测尿液中Trx、REG3G和AZGP1蛋白的表达水平。结果 照射后1 d,三种蛋白表达水平均升高,且与照射剂量有关。Trx蛋白5 Gy组照射后1 d较0 Gy组表达增高,8 Gy组照射后3 d、5 d较0 Gy组表达增高,差异具有统计学意义(P<0.05)。REG3G蛋白5 Gy组照射后3 d较0 Gy组表达增高,8 Gy组照射后5 d较0 Gy组表达增高,差异具有统计学意义(P<0.05)。AZGP1蛋白5 Gy组照射后1 d较0 Gy组表达增高,差异具有统计学意义(P<0.05)。结论 大鼠尿液中Trx和REG3G蛋白在全身照射后特定时间点有较为良好的剂量-效应关系。 |

Abstract: Objective To study the expression levels of thioredoxin (Trx), regenerating islet-derived 3 gamma (REG3G) and zinc-alpha2-glycoprotein (AZGP1) in urine of 60Co γ-ray irradiated rats. Methods According to the irradiation dosage, 30 Sprague Dawley (SD) rats were randomly divided into six groups (5 rats each group), then the rats were irradiated with 0, 0.5, 1, 3, 5, 8 Gy of 60Co γ-rays, respectively, at a dose rate of 1.0 Gy/min. The urines were collected at 1, 3, and 5 days after irradiation, respectively, to detect the expression levels of Trx, REG3G and AZGP1 proteins in the urine by ELISA. Results The results showed that 1 day after irradiation, the expression levels of three proteins all increased with the dose, The expression of Trx protein in 5 Gy group was higher than that of the 0 Gy group at the 3rd day after irradiation, its expression in 8 Gy group was all higher than that of the control group at the 3rd and 5th day after exposure, the differences were statistically significant (P<0.05). The expression of REG3G protein in 5 Gy group was higher than that of 0 Gy group at the 3rd day after exposure, and the 8 Gy group also so did at the 5th day after irradiation, the differences were statistically significant (P<0.05), too. The expression of AZGP1 protein in 5 Gy group was higher than that of 0 Gy group as well at the 1st day after irridiation (P<0.05). Conclusion The results suggested that Trx protein and REG3G protein in rat urine showed some good dose-response relationship at a specific time point in the urine of whole-body irradiated rats. |
